Sobre a tese
This research investigates how intestinal fibrosis develops in inflammatory bowel disease, where chronic inflammation produces obstructive scarring. The work demonstrates that activating intestinal fibroblasts via inflammatory signals does not always cause fibrosis, but TGF-β signaling acts as a central driver. In human intestinal organoids, fibrotic and inflammatory cues induced reversible epithelial changes, which were prevented by blocking TGF-β-related pathways. Furthermore, the medications upadacitinib and pirfenidone reduced expression of IL13RA2, a receptor linked to inflamed tissue and activated fibroblasts. Overall, the findings indicate that fibrosis arises from multiple cellular processes, suggesting that treatment requires targeting both the intestinal epithelium and fibroblast activation, potentially by repurposing existing drugs.
